Careful preparation of cell samples is the first crucial step towards reliable flow cytometric analysis. Suspensions of single, viable cells are a prerequisite for obtaining optimal results. In this technical presentation we will discuss how to prepare samples from diverse specimens like cultured cells, blood, or tissue. This includes gentle dissociation of tissues, removal of unwanted cells that could disturb the analysis, and getting reliable results from difficult samples. Based on various examples we will provide general guidelines, numerous tips and tricks, and straightforward troubleshooting.
