Summary:
Analysis of the mammalian cell cycle was one of the first applications to which flow cytometry was put in the 1960s. To assess cell cycle is relatively simple – we just stain the DNA of the cell with a fluorescent marker and measure the amount of fluorescence. However there are many things that need to be considered – to get the dye into the cell, it must be fixed or permeabilised so which protocol to use? Can I stain the DNA of live cells so they can be sorted according to cell cycle phase? There are a great many DNA-binding dyes available – which one should you use? How is the cytometer set up to ensure accurate measurement of the DNA in single cells? How should the DNA histogram produced be analysed – manually or automatically? What are the advantages and disadvantages of single parameter DNA analysis?
This webinar will assess the methods used for those who wish to take the first steps in DNA analysis and there will be a particular emphasis on the practical aspects of this type of analysis.
