Dear all,

would anyone have any good experience with successful use of a specific clone for the following: Brilliant Violet 421 anti-human IL17A antibody?

I notice it is on biolegend BL168 is the clone is one of the clones available.

I am trying to stain CD3+CD4+ Healthy human lymphocytes with this intracellularly.

any help would REALLY be appreciated.

best wishes,

Fiona.

I used this ab extensively on PBMC and we reported our results here: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC4688084/ - see Fig 6F. Works beautifully. We used eBio (now TF) fix/perm kit for fix/perm, and the ab at 1:50.

Caution - stim with Dynabeads did not work well, we figured that most of IL-17A producing cells are attaching to beads and we lose them when washing beads out. Plate-bound CD3 and soluble CD28 works nicely.

If you need more details, email me at belkina@bu.edu.

Hi Fiona, you might want to check out BenchSci. If you enter in BL168, you'll see that PE is the most published conjugate for that clone (of the cases where their AI recognized something).
